Descrizione: C9 is a plasma protein synthesized in the liver and monocytes consisting of a single polypeptide chain. C9 is a part of the membrane attack complex (MAC), an important component of the immune system. The MAC forms upon complement system activation by invading pathogenic bacteria and consists of the four major complement proteins: C5b, C6, C7 and C8. These complement proteins bind to the outer surface of the plasma membrane of the invading cell. C9 binds to the membrane associated C5b-8 protein, which leads to the circular polymerization of 12-18 C9 molecules. These polymerized C9 molecules form a ring structure in the membrane. Molecules can then diffuse freely through this transmembrane channel, causing cell lysis and destruction of the invading bacterial cell.

Descrizione: TOCA-1 is a 605 amino acid protein that localizes to the cytoplasm and the cytoskeleton, as well as to cytoplasmic vesicles and the cell membrane, and contains one FCH domain, one REM repeat and one SH3 domain. Existing as multiple alternatively spliced isoforms, TOCA-1 interacts with CDC42 and is required for the coordination of membrane tubulation with Actin cytoskeletal reorganization during endocytosis. Additionally, TOCA-1 is involved in membrane invagination, tubule formation and Actin polymerization. The gene encoding TOCA-1 maps to human chromosome 1, which spans 260 million base pairs, contains over 3,000 genes and comprises nearly 8% of the human genome.

Descrizione: ERCs (ERC1 and ERC2) also referred to as ELKS and CAST are related proteins which share an identical C-terminal sequence. They interact with the conserved RIM PDZ domain via an unusual PDZ binding motif. ERC2/CAST 1 is only expressed as a single RIM binding variant. All ERCs have been shown to interact with Rab6, a protein involved in membrane trafficking at the Golgi complex. The function of these proteins has not been determined yet. They may link Rab6 mediated non-neuronal membrane traffic at the Golgi complex to neuronal membrane traffic at the active zone executed via RIMs.

Descrizione: Probable core component of the endosomal sorting required for transport complex III (ESCRT-III) which is involved in multivesicular bodies (MVBs) formation and sorting of endosomal cargo proteins into MVBs. MVBs contain intraluminal vesicles (ILVs) that are generated by invagination and scission from the limiting membrane of the endosome and mostly are delivered to lysosomes enabling degradation of membrane proteins, such as stimulated growth factor receptors, lysosomal enzymes and lipids. The MVB pathway appears to require the sequential function of ESCRT-O, -I,-II and -III complexes. ESCRT-III proteins mostly dissociate from the invaginating membrane before the ILV is released. The ESCRT machinery also functions in topologically equivalent membrane fission events, such as the terminal stages of cytokinesis and the budding of enveloped viruses (HIV-1 and other lentiviruses).
