Descrizione: This antibody cocktail recognizes three melanoma-specific proteins, which include MART-1, Tyrosinase and gp100. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. The function of gp100 is not known but it is reported to be a useful marker for melanocytes and melanomas. This cocktail of three markers is designed for extremely sensitive labeling of formalin-fixed, paraffin-embedded melanomas and other tumors showing melanocytic differentiation.

Descrizione: This antibody recognizes a 66 kDa protein, which is identified as Tripartite motif-containing protein 29 (TRIM29). It interacts with the intermediate filament protein vimentin, a substrate for the PKC family of protein kinases, and with hPKCI-1, an inhibitor of the PKCs. TRIM29 protein contains both zinc finger and leucine zipper motifs, suggesting that the it may form homodimers and possibly associate with DNA. High expression of TRIM29 has been reported in gastric cancer and pancreatic cancer, and correlates with enhanced tumor growth and lymph node metastasis. TRIM29 is also able to distinguish lung squamous cell carcinoma from lung adenocarcinoma with ~90% positive accuracy, when used in a panel with TTF-1, p63, CK5/6, and Napsin-A antibodies.

Human malignant neoplasms carry rejection antigens that are recognized by the patients' autologous, tumor directed and specific, cytolytic, CD8 T lymphocyte clones (CTL). The MAGE family of genes codes an important group of antigens. It was identified that melanomas and primary glial brain tumors express common melanoma associated antigens (MAAs). Because MAGE-1 is expressed on a significant proportion of human neoplasms of various histological types (melanoma, brain tumors of glial origin, neuroblastoma, non-small cell lung cancer, breast, gastric, colorectal, ovarian, renal cell carcinomas) and not on normal tissues, the encoded antigen may serve as a marker of early detection and target for cancer immunotherapy.

Descrizione: This MAb is specific to lambda light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the lambda light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. It is important in memory formation and synaptic plasticity in the hippocampus. CD47 may play a role in membrane transport and/or integrin dependent signal transduction.

CD30 is synthesized as a 90 kDa precursor, which is processed in the Golgi complex into a membrane-bound phosphorylated mature 105/120 kDa glycoprotein. In Hodgkin's disease, CD30/Ki-1 antigen is expressed by mononuclear-Hodgkin and multinucleated Reed-Sternberg cells. It is also expressed by the tumor cells of a majority of anaplastic large cell lymphomas as well as by a varying proportion of activated T and B cells. This MAb distinguishes large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and lymphomas derived from resting and precursor lymphoid cells or from anaplastic carcinomas. About one third of the Ki-1 positive lymphomas lack the leukocyte common antigen (CD45).

Descrizione: CD46 acts as a cofactor for complement factor I, a serine protease, which protects autologous cells against complement-mediated injury by cleaving C3b and C4b deposited on host tissue. It may be involved in the fusion of the spermatozoa with the oocyte during fertilization. CD46 acts as a co-stimulatory factor for T-cells, which induces the differentiation of CD4 into T-regulatory 1 cells. T-regulatory 1 cells suppress immune responses by secreting interleukin-10, and therefore are thought to prevent autoimmunity. A number of viral and bacterial pathogens seem to exploit this property and directly induce an immunosuppressive phenotype in T-cells by binding to CD46.
