PAP-2 proteins function independently of Mg2+ and are insensitive to NEM (N-ethylmaleimide) inhibition. The lipid phosphates degraded by this family include ceramide 1-phosphate (C1P), sphingosine 1-phosphate (S1P), phosphatidic acid (PA) and lysophosphatidic acid (LPA). LPPR4 (lipid phosphate phosphatase-related protein type 4), also known as LPR4, PHP1, PRG1 or PRG-1, is a 763 amino acid multi-pass membrane protein that belongs to the PA-phosphatase related phosphoesterase family. Exclusively expressed in neurons, LPPR4 hydrolyzes lysophosphatidic acid (LPA) and facilitates axonal outgrowth during development and regenerative sprouting. LPPR4 exists as two alternatively spliced isoforms and is encoded by a gene located on human chromosome 1p21.2.

Descrizione: Stearoyl-CoA desaturase (SCD) is a regulatory enzyme in lipogenesis. The enzyme is constitutively expressed in adipose tissue and is induced in liver in response to fat free, high carbohydrate diet. Several SCD isoforms (SCD1, -2, and -3) exist in the mouse. Mice with a targeted disruption of the SCD1 isoform had reduced body adiposity, increased insulin sensitivity, and resistance to diet-induced weight gain. The protection from obesity involved increased energy expenditure and increased oxygen consumption. SCD1 deficiency shows activation of lipid oxidation in addition to reduced triglyceride synthesis and storage.

Descrizione: Stearoyl-CoA desaturase (SCD; EC 1.14.99.5) is an integral membrane protein of the endoplasmic reticulum that catalyzes the formation of monounsaturated fatty acids from saturated fatty acids. SCD may be a key regulator of energy metabolism with a role in obesity and dislipidemia. Four SCD isoforms, Scd1 through Scd4, have been identified in mouse. In contrast, only 2 SCD isoforms, SCD1 (MIM 604031) and SCD5, have been identified in human. SCD1 shares about 85% amino acid identity with all 4 mouse SCD isoforms, as well as with rat Scd1 and Scd2. In contrast, SCD5 shares limited homology with the rodent SCDs and appears to be unique to primates (Wang et al., 2005 [PubMed 15907797]).

Descrizione: ASAH2 Antibody: Sphingolipids are hydrolyzed by ceramidases to yield sphingosine and fatty acids. These ceramidases are classified according to the pH range that supports their optimal activity. ASAH2 is a neutral ceramidase and key regulator of sphingolipid signaling metabolites at the cell surface, catalyzing the hydrolysis of the N-acyl linkage of ceramide at an optimal pH of 6.5-8.5. ASAH2 is a type II integral membrane protein that can be cleaved to yield a soluble secreted protein and acts as a repressor of apoptosis both by reducing C16-ceramide, thereby preventing ceramide-induced apoptosis, and generating sphingosine. Sphingosine exerts both mitogenic and apoptosis-inducing activities, and its phosphorylated form functions as an intra- and intercellular second messenger. ASAH2 is ubiquitously expressed primarily expressed with higher levels in the intestine, kidney, skeletal muscle and heart. Recent studies indicate that ASAH2 encoded neutral ceramidase is a key enzyme for the catabolism of dietary sphingolipids and regulates the levels of bioactive sphingolipid metabolites in the intestinal tract.
