Descrizione: Protein kinases comprise a large group of encoded factors that regulate cellular processes by catalyzing the transfer of a phosphate group to a hydroxyl acceptor in serine, threonine or tyrosine residues (1,2). Kinases are capable of influencing the oncogenic potential of cell sytems at the level of oncoprotein or tumor suppressor protein phosphorylation states (1,2). STAP-2 is a protein that contains a pleckstrin homology (PH) domain and an SH2 domain, and associates with BRK (3). BRK (breast tumor kinase, Sik) is a 451 amino acid, nonreceptor protein-tyrosine kinase that is overexpressed in breast tumors and metastatic melanoma cell lines (4). Similar to the Src family of intracellular kinses, BRK is comprised of an SH3 domain, an SH2 domain, and a catalytic domain (5). STAP-2 is susceptiple to tyrosine phosphorylation and may be invovled in tyrosine kinase-mediated signaling cascades, whose aberrant function may lead to metastis (3).

Descrizione: Sam 68 is a protein that is phosphorylated on tyrosine and functions as a substrate for Src family tyrosine kinases during mitosis. Sam 68 also associates with several SH2 and SH3 domain-containing signaling proteins, such as GRB2 and PLC ?. Originally cloned as Ras GAP-associated p62, further investigations have shown that Sam 68 and Ras GAP-associated p62 are not antigenically related, nor are they encoded by the same gene. Like Sam 68, the Sam 68-like mammalian proteins, SLM-1 and SLM-2, demonstrate RNA binding activity. Also like Sam 68, SLM-1 is tyrosine phosphorylated and functions as an adapter protein for signaling molecules, including GRB2, PLC ?, Fyn and Ras GAP. SLM-2 is not tyrosine phosphorylated, nor does it appear to associate with GRB2, PLC ?, Fyn or Ras GAP, indicating that SLM-2 may not be an adapter protein for these proteins.

Descrizione: Nck is a highly conserved, oncogenic protein. It is a common target for the action of different surface receptors, encoding one SH2 and three SH3 domains, the Src homology motifs found in nonreceptor tyrosine kinases, Ras GTPase activating protein, phosphatidylinositol 3 kinase, and phospholipase Cg. Nck is widely expressed in various tissues and in cell lines from human, murine, and rat origins. Nck is phosphorylated on tyrosine, serine, and threonine residues in response to stimulation of EGF and PDGF in A431 and NIH 3T3 cells respectively. Like other SH2 containing proteins, Nck is associated with tyrosine autophosphorylated EGF or PDGF receptors via its SH2 domain. Overexpression of Nck leads to transformation of NIH 3T3 cells.
