Descrizione: <i>Pfu</i> DNA Polymerase, derived from the hyperthermophilic archae <i>Pyrococcus furiosus</i>, has superior thermostability and proofreading properties compared to other thermostable polymerases. It has a molecular weight of 90 kDa and can amplify DNA targets up to 2 kb. The elongation velocity is 0,2~0,4 kb/min (70~75 °C). <i>Pfu</i> DNA Polymerase possesses 3' to 5' exonuclease proofreading activity that enables the polymerase to correct nucleotide misincorporation errors. This means that<i> Pfu</i> DNA Polymerase-generated PCR fragments will have fewer errors than <i>Taq</i>-generated PCR inserts. Using <i>Pfu</i> DNA Polymerase results in blunt-ended PCR products, which are ideal for cloning into blunt-ended vectors. <i>Pfu</i> DNA Polymerase is superior for techniques that require high-fidelity DNA synthesis.

Descrizione: The Gram staining method was first described in 1844 by the Danish bacteriologist Hans Christian Gram, after whom the test was named. The Gram staining test for bacteria is one of the most important tests in microbiology and is often one of the first tests performed in the identification of bacteria. The Gram staining method utilises the properties of bacterial cell walls and the stain crystal violet. Gram-positive microorganisms have a higher peptidoglycan and lower lipid content than Gram-negative bacteria in their cell walls and this makes them capable of retaining the Gram stain.
