Descrizione: ATP-binding cassette (ABC) transporters are an evolutionarily conserved family of widely-expressed proteins that use ATP hydrolysis to catalyse the transport of various molecules across extracellular and intracellular membranes. Eukaryotic ABC transporters are largely responsible for trafficking hydrophobic compounds either within the cell as part of a metabolic process, outside the cell for transport to other organs, or for secretion from the body. The cholesterol-responsive transporter, ABCA7, maps to human chromosome 19 and mouse chromosome 10 and has been reported as a candidate regulator of ceramide transport in epidermal lipid reorganization. High expression levels of ABCA7 have been reported in myelolymphatic tissues, reticuloendothelial cells, peripheral leukocytes, thymus, spleen and bone marrow. This expression pattern of the two alternatively-spliced isoforms also indicates an involvement in lipid homeostasis in cells of the immune system, though the complete role of ABCA7 is not yet known. Full-length type I ABCA7 has shown plasma membrane localisation, while the type II splicing variant has shown expression predominantly in the endoplasmic reticulum.

C9 is a part of the membrane attack complex (MAC), an important component of the immune system. The MAC forms upon complement system activation by invading pathogenic bacteria and consists of the four major complement proteins: C5b, C6, C7 and C8. These complement proteins bind to the outer surface of the plasma membrane of the invading cell. C9 binds to the membrane associated C5b-8 protein, which leads to the circular polymerisation of 12-18 C9 molecules. These polymerised C9 molecules form a ring structure in the membrane. Molecules can then diffuse freely through this transmembrane channel, causing cell lysis and destruction of the invading bacterial cell.

Descrizione: LASS6 Antibody: The LASS (longevity assurance homolog) family members represent a subgroup of the homeobox gene family and are highly conserved from yeasts to mammals. Six members of this family of proteins have been characterized (LASS1-6) and all are involved in ceramide synthesis during cell growth regulation and cancer differentiation. Like the highly homologous LASS5, LASS6 is also an endoplasmic reticulum, multi-pass membrane protein. LASS6 is also involved in the synthesis of C14, C16 and C18-ceramide, but shows a preference for unsaturated fatty acids. LASS6 is broadly expressed in a wide range of tissues and microarray data suggests that it may play a role in cancer differentiation and early embryonic development.

Descrizione: SGMS2 is a bidirectional lipid cholinephosphotransferase capable of converting phosphatidylcholine (PC) and ceramide to sphingomyelin (SM) and diacylglycerol (DAG) and vice versa. Direction is dependent on the relative concentrations of DAG and ceramide as phosphocholine acceptors. SGMS2 directly and specifically recognizes the choline head group on the substrate. SGMS2 also requires two fatty chains on the choline-P donor molecule in order to be recognized efficiently as a substrate. SGMS2 does not function strictly as a SM synthase. SGMS2 is required for cell growth.Sphingomyelin (SM) is a major component of plasma membranes. It is preferentially concentrated in the outer leaflet and has a role in the formation of lipid rafts. SM synthases (EC 2.7.8.27), such as SGMS2, produce SM in the lumen of the Golgi and on the cell surface through the transfer of phosphocholine from phosphatidylcholine onto ceramide, yielding diacylglycerol as a side product (Huitema et al., 2004 [PubMed 14685263]).

CARD12 is a member of the Ced4 family and can induce apoptosis.

Descrizione: ASAH2 Antibody: Sphingolipids are hydrolyzed by ceramidases to yield sphingosine and fatty acids. These ceramidases are classified according to the pH range that supports their optimal activity. ASAH2 is a neutral ceramidase and key regulator of sphingolipid signaling metabolites at the cell surface, catalyzing the hydrolysis of the N-acyl linkage of ceramide at an optimal pH of 6.5-8.5. ASAH2 is a type II integral membrane protein that can be cleaved to yield a soluble secreted protein and acts as a repressor of apoptosis both by reducing C16-ceramide, thereby preventing ceramide-induced apoptosis, and generating sphingosine. Sphingosine exerts both mitogenic and apoptosis-inducing activities, and its phosphorylated form functions as an intra- and intercellular second messenger. ASAH2 is ubiquitously expressed primarily expressed with higher levels in the intestine, kidney, skeletal muscle and heart. Recent studies indicate that ASAH2 encoded neutral ceramidase is a key enzyme for the catabolism of dietary sphingolipids and regulates the levels of bioactive sphingolipid metabolites in the intestinal tract.
