Descrizione: High Mobility Group Protein B2 (HMGB2) belongs to the non-histone chromosomal high-mobility group protein family. Members of this family are chromatin-associated and widely spread in the nucleus of higher eukaryotic cells. HMGB2 contains 2 HMG box DNA-binding domains. It is associated with chromatin and has the ability to bend DNA, preferentially single-stranded DNA. It is shown that HMGB2 is able to efficiently bend DNA and form DNA circles. In addition, HMGB2 is involved in the final ligation step in DNA end-joining processes of DNA double-strand breaks repair and V(D)J recombination.

Descrizione: Interleukin 6 (IL-6) is a pleiotropic alpha-helical cytokine that plays important roles in acute phase reactions, inflammation, hematopoiesis, bone metabolism and cancer progression. IL-6 activity is essential for the transition from acute inflammation to either acquired immunity or chronic inflammatory disease. It is secreted by multiple cell types as a 22kDa-28kDa phosphorylated and variably glycosylated molecule. IL-6 induces signallling through a cell surface heterodimeric receptor complex composed of a ligand binding subunit (IL-6R) and a signal transducing subunit (gp130). IL-6 is a key factor for the growth of plasma cells.

Descrizione: Interleukin-4 (IL-4) is a cytokine produced by type 2 helper T cells, the Th2 cells. These cells tends to make a specific set of lymphokines including IL-4, IL-5, IL-6, IL-10, IL-13, IL-3 and GM-CSF and fail to produce IL-2, IFN-gamma, and lymphotoxin (TNF-beta). In addition, mast cells can produce IL-4. IL-4 exerts numerous effects on various hematopoietic cell types. On B cells, IL-4 promotes immunoglobulin class switching to IgE and IgG1 isotypes and upregulates MHC class II and CD23 expression. IL-4 promotes survival, growth, and differentiation of both T and B lymphocytes, mast cells and endothelial cells. In addition, IL-4 inhibits the production of TNF, IL-1, and IL-6 by macrophages.

Descrizione: CD152 (CTLA-4) and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally
homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an
intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulfide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by
cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+
T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation.
Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.

Descrizione: Human CD83 is a 40-50 kDa member of the Siglec (or sialic-acid-binding immunoglobulin-like lectin) family of transmembrane proteins. CD83 is a primary marker for dendritic cells. It is also found on B cells, neutrophils, monocytes and macrophages. Except for dendritic cells, CD83 expression is often transient. CD83 binds to sialic acids on target cells. Membrane CD83 promotes T cell proliferation, particularly of CD8+ cytotoxic T cells. Soluble CD83 is immunosuppressive and blocks T cell activation. On monocytes, CD83 is suggested to drive monocytes into a fibrocyte phenotype. A lack of membrane-expressed CD83 leads to an unusual IL-4/ IL-10 producing CD4+ T cell phenotype.
