Descrizione: GBE1 is a 702 amino acid protein that is expressed at high levels in muscle and liver and is involved in glycogen biosynthesis. Existing as a monomer, GBE1 catalyzes the transfer of alpha-1,4-linked glucosyl units from the outer end of a glycogen chain to an alpha-1,6 position on a neighboring glycogen chain and, via this catalytic activity, plays an essential role in glycogen accumulation. Defects in the gene encoding GBE1 are the cause of glycogen storage disease type 4 (GSD4) and adult polyglucosan body disease (APBD), the first of which is a metabolic disorder that is associated with the accumulation of polysaccharides and is characterized by liver disease during childhood. Unlike GSD4, APBD is a late-onset disorder that affects the central and peripheral nervous systems and is characterized by cognitive impairment, pyramidal tetraparesis and peripheral neuropathy.

Descrizione: Arginase I which is expressed almost exclusively in the liver, catalyzes the conversion of arginine to ornithine and urea . The human arginase I gene, which maps to chromosome 6q23, encodes a 322 amino acid protein. Arginase I exists as a homotrimeric protein and contains a binuclear manganese cluster. Arginase II catalyzes the same reaction as arginase I, but differs in its tissue specificity and subcellular location. Specifically, arginase II localises to the mitochondria. Arginase II is expressed in non-hepatic tissues, with the highest levels of expression in the kidneys, but, unlike arginase I, is not expressed in liver. The human arginase II gene, which maps to chromosome 14q24.1-q24.3, encodes a 354 amino acid protein. In addition, arginase II contains a putative amino-terminal mitochondrial localisation sequence.

Descrizione: GOT1L1, Glutamate oxaloacetate transaminase 1-like protein 1, is a 421 amino acid member of the class-I pyridoxal-phosphate-dependent aminotransferase family. Similar to glutamate-oxaloacetate transaminase (GOT1), GOT1L1 is found primarily as a homodimer in the cytoplasmic space but also has mitochondrial and chloroplastic isozymes. GOT1L1 transaminates 2-oxoglutarate with L-aspartate to yield oxaloacetate and L-glutamate. This reaction requires a pyridoxal phosphate cofactor to occur. The GOT1L1 peptidase is predominately expressed in the liver and serum levels of this protein can be used as an indicator of liver disease. Also, elevated glutamate concentrations in the brain interstitial fluids can lead to pathological brain conditions. The glutamate-scavenging properties of these aminotranferase type enzymes likely prevent glutamate excitotoxicity and the long-lasting neurological deficits seen after stroke.

Descrizione: GSTZ1 is a member of the glutathione S transferase (GSTs) super family, encoding multifunctional enzymes important in the detoxification of electrophilic molecules, including carcinogens, mutagens, and several therapeutic drugs, by conjugation with glutathione. This enzyme also plays a significant role in the catabolism of phenylalanine and tyrosine. Several transcript variants of this gene encode multiple protein isoforms. GSTZ1 shows minimal glutathione-conjugating activity with ethacrynic acid and 7-chloro-4-nitrobenz-2-oxa-1,3-diazole and maleylacetoacetate isomerase activity. It has low glutathione peroxidase activity with T butyl and cumene hydroperoxides and is able to catalyze the glutathione dependent oxygenation of dichloroacetic acid to glyoxylic acid. Highest expression in liver followed by kidney, skeletal muscle and brain. Also expressed in melanocytes, synovium, placenta, breast and fetal liver and heart.
