Descrizione: This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and catalyzes the last steps of estrogen biosynthesis, three successive hydroxylations of the A ring of androgens. Mutations in this gene can result in either increased or decreased aromatase activity; the associated phenotypes suggest that estrogen functions both as a sex steroid hormone and in growth or differentiation. Descrizione: Apolipoprotein E (ApoE) is a lipoprotein involved in fat metabolism and acts as cholesterol carrier between cells and across tissues. On a genetic level, three APOE alleles are described, APOE2, APOE3 and APOE4. These alleles give rise to six APOE isoforms, which are differentially implicated in various diseases. In the peripheral system, APOE4 is linked to increased risk of atherosclerosis. In the CNS, the ability of APOE4 in clearing beta-amyloid is impaired, while APOE3 and APOE2 are more efficient in performing this task. The APOE4 genotype in particular has been linked to increased risk for developing Alzheimer's Disease.

Descrizione: Cavin 1-PTRF has been shown to be an essential component of caveolae. Cavin 1-PTRF appear to be recruited to membrane domains containing PS, cholesterol and oligomerized caveolins. Binding of Cavin 1-PTRF to these domains stabilizes the membrane curvature to produce the classic flask shape of caveolae. Disassembly of Cavin 1- PTRF releases caveolar components, including caveolins into the plasma membrane. Succinctly it appears that Cavin 1-PTRF regulates caveola structure and function.

Descrizione: NPC2 is a secreted protein mapping against gene 14q24.3 (1,2). NPC2 regulates the lipid composition of sperm membranes during maturation in the epididymis(1,2). Mutations in the NPC2 gene may cause Nieman-Pick type C2 disease and frontal lobe atrophy (1,2,3). Nieman-Pick type C2 is a fatal hereditary disease characterized by defective lysosome release of cholesterol (3). The disease is caused by HE1 deficiency, a lysosmal protein proven to be undetectable in fibroblasts from NPC2 patients (3). This differentiates NPC2 from NPC1, as NPC1 has HE1 protein present (3).

INSIG1 binds to the sterol-sensing domains of SREBP cleavage-activating protein (SCAP) and HMG CoA reductase, and is essential for the sterol-mediated trafficking of the two proteins.The protein encoded by this gene is highly similar to the protein product encoded by gene INSIG1. Both INSIG1 protein and this protein are endoplasmic reticulum proteins that block the processing of sterol regulatory element binding proteins (SREBPs) by binding to SREBP cleavage-activating protein (SCAP), and thus prevent SCAP from escorting SREBPs to the Golgi.
