Descrizione: Insulin-like growth factors (IGFs) comprise a family of endocrine, paracrine and autocrine polypeptides consisting of the ligands IGF1 and IGF2, two receptors (IGF1R, IGF2R), at least 6 IGF-binding proteins (IGFBPs) and IGFBP proteases. Among the binding proteins, IGFBP6 is unique because of its N-terminal disulphide linkages and its marked binding preference for IGF2. It is a potent inhibitor of the interaction between IGF2 and its receptor IGF1R, thus preventing major functions of IGF2, such as induction of proliferation, differentiation, cell adhesion, or colony formation. In particular, IGFBP-6 inhibited the growth of neuroblastoma and rhabdomyosarcoma xenografts. GFBP-6 is expressed in many tissues, including lung, liver, gut and the central nervous system.

Descrizione: Interleukin-12 subunit beta (IL-12B) belongs to the type I cytokine receptor family. It contains 1 fibronectin type-III domain and 1 Ig-like C2-type domain. IL-12B is a cytokine that acts on T and natural killer cells, and has a broad array of biological activities. Descrizione: Below 35 to 40 °C gelatin swells and absorbs approximately 5 to 10 times its weight of water to form a gel. Gelatin is soluble in glycerol and acetic acid, and more soluble in hot than in cold water. It is practically insoluble in most organic solvents such as alcohol, chloroform, carbon disulphide, carbon tetrachloride, ether, benzene, acetone and oils. Sterile solutions of gelatin stored cold are stable indefinitely, but at elevated temperatures hydrolysis or rupture of peptide bonds occurs, increasing the number of free amino groups. Gel strength and viscosity gradually weaken upon prolonged heating in solution above ~40 °C; this degradation is accelerated by extremes in pH, proteolytic enzymes and bacterial action.

Descrizione: The IL12 receptor complex, formed by IL12RB1 and IL12RB2, mediates the type I immune responses of various types of lymphocytes. Its ligand, IL12, is a heterodimeric cytokine composed of IL-12p35 and IL-12p40 subunits that are linked via disulphide bonds. Ligation of IL-12 to its receptor involves the binding of IL-12p35 to IL12RB1 and IL-12p40 to IL12RB2. This will result in the activation of tyrosine kinase 2 (TYK2), which is associated with the IL12RB1 chain and Janus kinase 2 (JAK2), which is associated with the IL12RB2 chain. Activated TYK2 and JAK2 direct the phosphorylation of STAT4. IL12RB1 is present on all lymphocytes, while the expression of IL12RB2 is tightly regulated. It has shown that the expression of IL12RB2 is limited to Th2 cells. IL12RB2 subunit plays an important role in Th1 cell differentiation, since its absence leads to an abortive Th1 differentiation that has dysfunctional production of Th1 effector molecules.

Descrizione: A disulphide-linked heterodimer, consisting of mb-1 (or CD79a) and B29 (or CD79b) polypeptides, is non-covalently associated with membrane-bound immunoglobulins on B cells. This complex of mb-1 and B29 polypeptides and immunoglobulin constitute the B cell Ag receptor. CD79a first appears at pre B cell stage, early in maturation, and persists until the plasma cell stage where it is found as an intracellular component. CD79a is found in the majority of acute leukemias of precursor B cell type, in B cell lines, B cell lymphomas, and in some myelomas. It is not present in myeloid or T cell lines. Anti-CD79a is generally used to complement anti-CD20 especially for mature B-cell lymphomas after treatment with RituximAb (anti-CD20). This antibody will stain many of the same lymphomas as anti-CD20, but also is more likely to stain B-lymphoblastic lymphoma/leukemia than is anti-CD20. Anti-CD79a also stains more cases of plasma cell myeloma and occasionally some types of endothelial cells as well.

Descrizione: CD152 and CD28, together with their ligands B7-1 and B7-2, constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD152 and CD28 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD152 and CD28 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD152 and CD28 are both expressed on the cell surface as disulphide-linked homodimers or as monomers. CD152 was originally identified as a gene that was specifically expressed by cytotoxic T lymphocytes. However, CD152 transcripts have since been found in both Th1 and Th2, and CD4+ and CD8+ T cell clones. Whereas, CD28 expression is constitutive on the surfaces of 95% of CD4+ T cells and 50% of CD8+ T cells and is down regulated upon T cell activation, CD152 expression is upregulated rapidly following T cell activation and peaks approximately 24 hours following activation. Although both CD152 and CD28 can bind to the same ligands, CD152 binds to B71 and B72 with 20-100-fold higher affinity than CD28.
