Descrizione: A Fluor-de-Lys® fluorescent assay system. The HDAC8 Fluorescent Activity Assay/Drug Discovery Kit is a complete assay system designed to measure the lysyl deacetylase activity of the recombinant human HDAC8 included in the kit. The kit is ideal for chemical library screening for candidate inhibitors or activators or kinetic assay of the enzyme under varying conditions. The Fluor de Lys® HDAC8 assay is based on the Fluor de Lys® Substrate and Fluor de Lys® Developer combination. The assay procedure has two steps. First, the Fluor de Lys® Substrate, which comprises an acetylated lysine side chain, is incubated with HDAC6. Deacetylation of the substrate sensitizes the substrate so that, in the second step, treatment with the Fluor de Lys®Developer produces a fluorophore.

Descrizione: MTT, XTT, and resazurin are reduced by mitochondrial metabolic activity to yield coloured or fluorescent products, and thus are useful for and assaying cell viability and quantitating cell number. MTT and XTT are reduced to coloured formazin salts that can be measured by absorbance. MTT generates an insoluble formazin salt, requiring cell lysis before the absorbance can be measured, while XTT does not require cell lysis for measurement. Resazurin is a non-fluorescent blue dye that is reduced to the pink fluorescent compound resorufin, which can be measured by fluorescence or absorbance.

In this assay, TdT enzyme catalyses the addition of labeled dUTP to the 3’ ends of cleaved DNA fragments. Fluorescent dye-conjugated dUTP can be used for direct detection of fragmented DNA by fluorescence microscopy or flow cytometry.
